Description

Phenol-Chloroform-Isoamyl alcohol mixture in the ratio of 25:24:1 (v/v/v), CAS # 136112-00-0, 67-66-3, 123-51-3, is a solution used in the purification of nucleic acids.

Phenol:Chloroform:Isoamyl alcohol 25:24:1 is used in the extraction of DNA for the generation of DNA minisatellite fingerprints. It is also used for purifying RNA.

This Thermo Scientific Chemicals brand product was originally part of the Alfa Aesar product portfolio. Some documentation and label information may refer to the legacy brand. The original Alfa Aesar product / item code or SKU reference has not changed as a part of the brand transition to Thermo Scientific Chemicals.

General Description

  • This solution contains a mixture of pure chloroform, isoamyl alcohol and phenol saturated with Tris-EDTA, with a stabilized pH of 8.0

Applications

  • Phenol-Chloroform-Isoamyl alcohol mixture solution (25:24:1) is used in the extraction of nucleic acids, DNA and RNA
  • When nucleic acids are extracted with the Phenol-Chloroform-Isoamyl alcohol mixture solution, a clear component separation is achieved. The proteins present in the sample are denatured and removed (collected in the organic phase or in the interphase), while nucleic acids remain in the aqueous phase. The chloroform denatures proteins and facilitates the separation of the aqueous and organic phases, and the isoamyl alcohol reduces foaming during extraction.
  • It is also used to generate DNA minisatellite fingerprints
