- Recognizes human PRL
- PRL (Prolactin) is a pleiotropic hormone mainly synthesized by lactotrophs of anterior pituitary gland
- Causes breasts to grow and make milk during pregnancy and after birth
- Normally high levels for pregnant women and new mothers
- Plays role in metabolism, regulation of immune system and pancreatic development
- Normally during pregnancy prolactin serum levels increased to hold insulin resistance
- Involved in regulation of glucose metabolism
Process:
- Antibody specific for PRL pre-coated onto microwells
- PRL protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for PRL is added to detect captured PRL protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
