The reaction buffer for Thermo Scientific Phire Hot Start II DNA Polymerase is available in three formats: non-colored version with and without detergents or Green version containing a density reagent and two tracking dyes for direct loading of PCR products on gel.

Thermo Scientific Phire Reaction Buffer is designed for Phire Hot Start II DNA Polymerase. This 5X buffer solution is available in three formats, a non-colored version with or without detergents and a green version that contains a density reagent and two tracking dyes for direct loading of PCR products on gel. The colored buffer does not interfere with enzyme performance and is compatible with downstream applications such as DNA sequencing, ligation, and restriction digestion.

Phire Reaction Green Buffer includes a blue die that migrates with 3–5 kb DNA fragments in 1% agarose gel and a yellow dye that migrates faster than 10 bp DNA fragments in 1% agarose gel. The dyes have excitation peaks at 424 nm and 615 nm.

The concentration of Mg2+ in Phire Reaction Buffer is optimized to work well for most amplicons. The buffer contains 1.5 mM MgCl2 at final reaction concentration. Separate tubes of MgCl2 and DMSO are provided for further optimization.

Notes

  • For applications that require PCR product analysis by absorbance or fluorescence excitation, the colorless Phire Reaction Buffer is recommended.
